    张劲松,博士,研究员,博士生导师

        1986年和1991年在北京大学生物系获学士和博士学位。1991-1994年,中科院遗传所助理研究员。1994-1997年,美国堪萨斯州立大学博士后研究。1997-2006年,中科院遗传发育所副研究员。2006年4月至今,中科院遗传发育所研究员。2009年获得国家杰出青年基金资助。2015年至今,中国科学院大学岗位教授。担任十四五国家重点研发计划首席。主持和/或参与了国家重点研发计划、国家自然科学基金重点和面上项目和中科院先导专项等。获得基因发明专利40余项,在Nature Commun、PNAS、Plant Cell和Mol Plant等杂志发表SCI论文100余篇。任中国作物学会大豆专业委员会理事,中国植物生理学会开花与衰老专业委员会理事。JIPB、aBIOTECH、Plant Hormones、大豆科学和植物生理学报等杂志编委。

    研究领域:

        大豆油脂积累和籽粒性状调控及水稻乙烯信号转导机制研究
        一、大豆油脂积累、籽粒性状和耐逆性调控的机制研究
        大豆是重要的粮油饲兼用作物,在国民经济中起着举足轻重的作用。但我国大豆需求80%以上依赖进口,因此提高大豆产量和油脂含量是国家重大战略需求。由于耕地面积有限,提高单位面积产量和油脂积累就是重要研究课题。
        利用大豆自然群体、遗传群体和突变群体,通过基因共表达调控网络、QTL和图位克隆及多组学技术方法,鉴定了一系列调控大豆油脂积累、籽粒性状和耐逆性的基因和/或模块(图1)。在大豆油脂调控方面,鉴定了大豆油脂积累的正调控转录因子基因包括MYB331NFYAZF351ZF392MYB73bZIP123DREBLDOF4/11,及油脂积累的负调控因子ERFA和负调控模块JAZ3-FVE-HDA。其中NFYA-ZF392-ZF351可形成模块促进油脂积累,而ERFA可通过与NFYA互作抑制下游基因bZIP123ZF392表达,ERFA-NFYA-bZIP123/ZF392模块在种子发育后期抑制油脂积累(Liu et al., 2025),调控其他储藏成分如蛋白的积累。也鉴定了JAZ3-FVE-HAD模块负调控大豆油脂积累。JAZ3可通过招募FVE和HAD抑制bZIP123对下游基因的激活作用,在籽粒发育后期抑制油脂积累,促进蛋白等其他储藏物质积累(图1)。
        通过正向遗传学方法鉴定了第一个大豆粒重调控基因PP2C-1,也鉴定了JAZ3、PLATZ和mR172a-ERF416/413模块等调控大豆籽粒大小和油脂含量。通过QTL和突变体分析鉴定了大豆粒重和株型等产量性状的新调控基因。也鉴定了一系列大豆耐盐耐旱调控新基因并揭示了其调控分子机制和网络。上述专利基因已提供11个单位使用。

        红色为大豆油脂调控、籽粒性状调控和近期鉴定的耐逆性调控蛋白。多数油脂调控蛋白也参与调控植物耐逆性。绿松石颜色为大豆油脂的负调控蛋白或负调控模块,在大豆籽粒发育后期起作用,可能促进蛋白积累。
        二、水稻乙烯信号转导及农艺性状调控机制研究
        水稻是重要的单子叶作物,主要生活在半水生环境中。乙烯在这个过程起着重要作用,但乙烯信号通路的研究在水稻中尚不十分清楚。水稻黄化苗的乙烯反应包括根生长的抑制和地上部分胚芽鞘伸长的促进(图2),这与双子叶植物拟南芥的三重乙烯反应不同。我们突破了水稻乙烯突变体鉴定瓶颈,通过筛选31000份T-DNA插入突变体库和25000份EMS突变体库,获得一系列水稻乙烯反应的突变体命名为“猫胡子”(mhz1-12)(图2)。除了保守的组分如乙烯受体、EIN2和EIL1/2以外,鉴定的新的乙烯信号转导调控基因如组氨酸激酶MHZ1、膜蛋白MHZ3、RNA结合蛋白MHZ9和GDSL脂酶MHZ11等,可分别通过组氨酸磷酸传递、与乙烯受体结合调控乙烯信号开关、调控翻译过程,以及通过调控膜甾醇含量影响膜流动性促进受体信号输出等方面调控水稻乙烯反应(图3)。近期也从大量水稻资源中筛选出乙烯反应改变的资源多份,鉴定了部分相关基因,其参与抗病和耐盐耐旱等过程,为育种家提供了重要基因资源。相关研究受到国内外乙烯生物学研究领域广泛关注和高度评价。

        野生型对照中,水稻黄化苗的根受到乙烯抑制而地上部胚芽鞘伸长受到促进。所有突变体的根对乙烯不敏感或钝感,而胚芽鞘呈现为不敏感、钝感或与对照类似表型。

        在上述调控途径中,MHZ1、MHZ3、MHZ9和MHZ11为新的乙烯途径调控组分。MHZ4、MHZ5、MHZ2、MHZ10和GY1是与其他激素包括ABA、生长素和JA途径互作的新节点。
